dc.subjectWe optimized the purification strategies of large macromolecular complexes in the hyperthermophile P. furiosus. This thesis describes the optimization we did, the biochemical strategies used and the data analysis performed. A major part of this thesis is focused on the crude purification methods, whereby the complexes were purified in silico after cryo-EM images were taken of grids containing a mixture of several macrocomplexes. Molecules of interest were the 50S ribosome and encapsulins.
