dc.description.abstract | The goal of this study is to design a research method to visualize the changes in expression of different enzymes and receptors involved in the lipolysis of dairy cows. To monitor the expression of 6 enzymes and/or receptors, ATGL; HSL; Perilipin; PPARy; PDE3b and GPR109A, involving the lipolysis during ketosis towards clinical signs, the lipid tissue of 2 bovine patients and 1 control cow were used to extract the RNA. The extracted RNA was used to create cDNA. By adding the primers and using the PCR-method, the expression of the 6 enzymes were visualized.
In contradiction to literature, the expression of ATGL seems to decrease during clinical ketosis. There is no logical explanation of the expression of HSL in this study. The expression in both patients is completely different during clinical ketosis. The expression of GPR109A, PPARy and perilipin also seem quite unclear in these two patients. The primers of PDE3b were ineffective.
There is not one clear factor which elucidates the development of clinical signs during ketosis. All except ATGL have a contradictory expression in the lipid tissue of both patients and the expression of ATGL is not aligned with other literature.
More research is needed to determine the factor which causes the differences in clinical ketosis between cows postpartum. | |
